MOTS-c is a 16-amino-acid peptide encoded not in the nuclear genome but inside mitochondrial DNA — a genuinely unusual origin. Discovered in 2015 at USC, it acts like a hormone that mitochondria send out to the rest of the body to coordinate metabolic response to stress.

Circulating MOTS-c levels drop with age and with insulin resistance. Restoring them in animal models improves glucose disposal, endurance capacity, and healthspan markers. It essentially mimics the effect of exercise at the cellular signaling level.
